The Waters XBridge Peptide BEH C18 OBD preparative column is designed for reversed-phase purification of peptides and protein-derived fragments. A 10 mm × 250 mm preparative bed contains 5 µm bridged-ethyl hybrid particles with 300 Å pores and C18 bonded chemistry, providing hydrophobic selectivity for peptide mixtures. The wide-pore format supports access by larger peptides and can be used when preparative fraction collection is required after peptide synthesis, digestion or characterization. Establish a gradient using solvents and modifiers compatible with peptide solubility, detection and downstream recovery. Evaluate loading capacity and purity independently because retention and resolution depend strongly on sequence, charge, hydrophobicity and mobile-phase conditions. The operating pH range is 1–12, with maximum pressure 6000 psi (415 bar). Equilibrate the column before sample loading, use appropriate preparative connections and flush it with a suitable solvent after the run to minimize precipitation or buildup.
