Agilent Bio SEC-5 Size Exclusion HPLC Column uses 5 µm fully porous packing with a nominal 100 Å pore size in a 7.8 mm internal diameter by 300 mm stainless steel column. Size exclusion chromatography separates dissolved biomolecules primarily by hydrodynamic size, with larger species eluting sooner because they access less of the particle pore volume. The 100 Å version is suited to suitable low-molecular-weight biomolecule and peptide separations within its effective pore range; it should not be assumed to offer the same fractionation window as 300 Å or 1000 Å SEC media. The longer 300 mm bed supports resolution in analytical SEC methods where sample loading and mobile-phase conditions are controlled. Use a compatible aqueous buffer, filter samples and minimize nonspecific interactions to maintain reproducibility. Select the column based on analyte size distribution rather than assuming all Bio SEC-5 versions are interchangeable.
