MycoAlert® PLUS Mycoplasma Detection Kit (30 Tests) is used for detecting viable mycoplasma contamination in cell-culture supernatants with a rapid bioluminescent assay. The workflow lyses mycoplasma in the sample and uses mycoplasmal enzyme activity to convert ADP to ATP; a luciferase reaction then converts the ATP signal into light. Compare the luminescence reading before and after substrate addition to determine whether mycoplasma is present. The PLUS formulation is suitable for less-sensitive plate luminometers and multifunctional readers and also works with tube luminometers. The assay requires two reagent additions and two luminescence readings and can produce a result in less than 20 minutes. It detects common Mollicutes associated with cell-culture contamination, including Mycoplasma, Acholeplasma, Entomoplasma and Spiroplasma, while Ureaplasma is outside the detection scope. The kit contains three 1.2 mL lyophilized reagent vials, three 1.2 mL lyophilized substrate vials and three 3 mL assay-buffer vials for 30 tests.
