Answer:It measures mycoplasmal enzyme activity that converts ADP to ATP and then quantifies the ATP through a luciferase luminescence reaction.
Answer:The assay can produce results in less than 20 minutes.
Answer:It is suitable for tube luminometers, plate luminometers and multifunctional readers, including less sensitive plate-reading systems.
MycoAlert® PLUS Mycoplasma Detection Kit, 100 Tests is used for rapid biochemical screening of viable mycoplasma contamination in cell-culture supernatants. The assay lyses contaminating mollicutes and measures enzymes that are widely present in mycoplasma but absent from eukaryotic cells. After the PLUS substrate is added, these enzymes convert ADP to ATP; a luciferase reaction then converts ATP into a light signal. Comparing the luminescence before and after substrate addition produces a B/A ratio that indicates whether contamination is present. The PLUS formulation generates stronger light output than the standard MycoAlert format, making it suitable for less sensitive plate luminometers and multifunctional readers. Results are available in less than 20 minutes using a simple two-reagent workflow. The assay detects common Mycoplasma, Acholeplasma, Entomoplasma and Spiroplasma contaminants, but does not detect Ureaplasma, and it is intended for routine cell-culture contamination monitoring.
