Description
Vedolizumab297.rMAb is a nontherapeutic recombinant monoclonal antibody (mAb) that specifically recognizes an extracellular epitope on Human Lymphocyte Peyer's patch adhesion molecule-1 (LPAM-1) like the Vedolizumab therapeutic antibody. Vedolizumab is a humanized therapeutic mAb that was made using the immunoglobulin (Ig) variable region complementary determining regions (CDRs) of the human LPAM-1-specific mouse ACT-1 mAb placed into the consensus framework region sequences of human IgG1, kappa. The Vedolizumab297.rMAb was engineered to incorporate an asparagine with alanine at position 297 (N27A) of the human IgG1 heavy chain to reduce potential nonspecific staining caused by Fc receptor binding. LPAM-1 is a heterodimer comprised of the noncovalently bound type I transmembrane integrin glycoproteins, 150 kDa Integrin α4 (aka, CD49d; encoded by ITGA4) and 130 kDa Integrin β7 (encoded by ITGB7) that form Integrin α4β7. LPAM-1 (Integrin α4β7) heterodimer is variably expressed on the cell surface of leukocytes including B lymphocytes and plasma cells, T lymphocytes, NK cells, and eosinophils. It acts as a homing receptor that facilitates the adhesion of leukocytes to the mucosal vascular addressin, MAdCAM-1 which is selectively expressed on endothelial venules of gut lamina propria venules and on high endothelial venules (HEV) within Peyer's patches and mesenteric lymph nodes. This interaction plays a major role in the migration of leukocytes into gut-associated lymphoid tissues. Additional ligands include fibronectin in the extracellular matrix and Vascular cell adhesion molecule 1 (VCAM-1, CD106) that is expressed on endothelial cells. LPAM-1 (Integrin α4β7) is involved in chronic inflammatory disorders of the gastrointestinal tract. The therapeutic Vedolizumab antibody can inhibit the migration of gut-homing memory T cells into the gastrointestinal submucosa by blocking the interaction between Integrin α4β7 and its ligand, the vascular addressin MAdCAM-1. Vedolizumab297.rMAb is intended for research use only and is not intended for use in therapeutic or diagnostic procedures for humans or animals.
BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.
The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to the dye under optimum conditions and unreacted dye was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

2. Alexa Fluor® 647 fluorochrome emission is collected at the same instrument settings as for allophycocyanin (APC).
3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
4. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
5. For fluorochrome spectra and suitable instrument settings, please refer to our Multicolor Flow Cytometry web page at www.bdbiosciences.com/colors.
6. An isotype control should be used at the same concentration as the antibody of interest.
