Applications of heparin affinity chromatography
Cytiva's heparin resin allows for fast and reliable separations of biomolecules with an affinity for heparin, including antithrombin III antibodies, coagulation factors, and other plasma proteins. DNA-binding proteins, lipoproteins, protein synthesis factors, enzymes that act on nucleic acids, and steroid receptors can also be purified.

Cytiva Heparin Sepharose 6 Fast Flow is an affinity chromatography resin for biomolecules that interact with immobilized heparin, including antithrombin and selected nucleic acid-binding proteins. The matrix comprises cross-linked 6% agarose beads carrying a covalently coupled porcine heparin ligand. Its average particle size is approximately 90 µm, supporting a mechanically stable chromatography bed for purification development and production workflows. The resin is supplied preswollen in 20% ethanol containing sodium acetate; remove the preservative and equilibrate thoroughly with binding buffer before sample application. Binding can involve specific affinity as well as electrostatic interactions, and increasing buffer salt concentration frequently releases retained proteins. Optimize pH, loading and salt-gradient conditions for the target biomolecule rather than assuming universal selectivity. The one-litre pack contains bulk resin for column packing, not a ready-to-use prepacked cartridge. Follow compatible storage and cleaning conditions to preserve ligand activity.
