Answer:The prepacked bed contains 5 mL of Ni Sepharose High Performance resin.
Answer:The bed is approximately 16 × 25 mm with a recommended flow rate of 5 mL/min.
Answer:It has five times the resin volume, supporting greater total protein loading when binding conditions are comparable.
Cytiva HisTrap HP 5 mL affinity columns provide a larger prepacked bed for preparative purification of polyhistidine-tagged proteins. Each column contains 5 mL of Ni Sepharose High Performance resin, a nickel-charged, highly cross-linked agarose medium with an average particle size of 34 µm. The bed measures approximately 16 × 25 mm and supports a recommended flow rate of 5 mL/min. At least 40 mg of His-tagged protein can bind per mL of medium under suitable conditions, making the larger bed useful when load mass exceeds the capacity of a 1 mL format. Apply clarified sample in an appropriate binding buffer, reduce nonspecific association with a controlled washing step, and recover target protein with a suitable imidazole-containing elution buffer. The prepacked format can connect to compatible syringe, pump and chromatography systems. Monitor pressure and flow, avoid exposing the resin to incompatible solutions, and maintain the prescribed storage medium between runs.
