Cytiva Capto S is a strong cation-exchange chromatography resin for capture and intermediate purification of proteins and other positively charged biomolecules. The functional sulfonate groups are coupled through dextran surface extenders on a highly cross-linked agarose matrix, providing access to binding sites while supporting high-flow packed-bed operation. The 5 L resin quantity supports development and production-scale column packing, where process requirements justify bulk chromatography media. Select a loading pH at which the target interacts with the negatively charged ligand, then evaluate conductivity, binding capacity, recovery and impurity clearance experimentally. A salt or pH gradient can be used for elution, subject to the process design. The manufacturer's recommended storage medium includes 20% ethanol and 0.2 M sodium acetate. Before use, replace storage liquid with the appropriate equilibration buffer and pack the column under controlled conditions. Confirm the column's packing quality, pressure-flow behavior and cleaning compatibility before qualification. Capto S is a cation exchanger, not an anion-exchange resin.
