Description
Easily and efficiently isolate highly purified mouse naïve CD4+ T cells (CD4+CD62L+) from single-cell suspensions of mouse splenocytes or other tissue samples using immunomagnetic negative selection followed by positive selection, with the EasySep™ Mouse CD4+CD62L+ T Cell Isolation Kit. Widely used in published research for more than 20 years, EasySep™ combines the specificity of monoclonal antibodies with the simplicity of a column-free magnetic system.
In this EasySep™ selection procedure, naïve CD4+ T cells are first pre-enriched by negative selection using the EasySep™ Mouse Naïve CD4+ T Cell Pre-Enrichment Cocktail containing biotinylated antibodies directed against non-naïve CD4+ T cells. Unwanted cells expressing the following markers are targeted for removal: CD8, CD11b, CD11c, CD19, CD24, CD25, CD45R, CD49b, TCRγ/δ, TER119. Then, CD4+CD62L+ cells, pre-labeled with CD62L PE, are selected using the EasySep™ PE Selection Cocktail. The desired cells are labeled with antibodies and magnetic particles. Labeled cells are separated using an EasySep™ magnet and by simply pouring off the unwanted cells. The cells of interest remain in the tube. Following magnetic cell isolation, the desired cells are ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. Note: The positively selected CD62L+ cells have already been PE-labeled, so their purity can be assessed directly by flow cytometry.
