Gibco DMEM/F-12 (1:1) is a liquid basal medium combining Dulbecco's Modified Eagle Medium and Ham's F-12 nutrient mixture for mammalian cell culture. The 500 mL formulation contains L-glutamine and phenol red and does not contain HEPES. Its broad nutrient composition supports culture systems including epithelial, glial, endothelial, and fibroblast cells when paired with the supplements required by each cell type. This is a basal medium rather than a complete growth-factor- or serum-supplemented formulation. Determine serum, growth-factor, antibiotic, or other additives from the validated culture protocol; use suitable CO₂ control because the bicarbonate-buffered formulation relies on an appropriate incubator atmosphere. Maintain sterile technique during dispensing, avoid repeated warming, and store the medium at 2–8 °C protected from light. Do not confuse it with DMEM/F-12 versions formulated without L-glutamine or with HEPES.
