Application Note

NuPAGE™ Antioxidant is a reducing-environment additive for NuPAGE protein electrophoresis and transfer workflows. It is added to the running buffer when samples have been reduced with NuPAGE Sample Reducing Agent or another compatible reducing reagent. During electrophoresis, the antioxidant migrates with the proteins and helps maintain sensitive amino-acid residues in a reduced state, limiting reoxidation of groups such as methionine- and tryptophan-associated oxidation-sensitive sites. This 15 mL configuration is optimized for the neutral-pH chemistry of NuPAGE Bis-Tris and related NuPAGE gel systems and is not intended as a universal additive for unrelated gel chemistries. Maintaining a reducing environment can improve consistency when analyzing proteins that contain redox-sensitive groups or disulfide-dependent structures. Store and handle the reagent under the conditions specified for NuPAGE electrophoresis reagents.
