Gibco Opti-MEM I Reduced Serum Medium is a sterile-filtered liquid medium designed for reduced-serum mammalian cell culture and lipid-based transfection. This formulation contains L-glutamine and phenol red, while fetal bovine serum is not supplied as part of the medium. It incorporates insulin, transferrin, hypoxanthine, thymidine, and trace elements to support cell growth under conditions using less added serum than standard medium. The 1× medium is supplied in a 500 mL bottle. It can be used with adherent and suspension mammalian cell types, including CHO, HEK, BHK-21, and related cells, and is compatible with Lipofectamine-based transfection workflows. Its sodium bicarbonate buffering system requires a CO2-controlled culture environment. For extended cell growth, serum supplementation should be determined for the cell line and experimental setup; the medium can also serve as a serum-free diluent during transfection complex formation. Store at 2–8 °C protected from light. The shelf life is 18 months from manufacture.
