Gibco Opti-MEM I Reduced Serum Medium without phenol red is a sterile-filtered mammalian cell culture and transfection medium for experiments where a phenol-red-free formulation is preferred. It retains L-glutamine and the reduced-serum Opti-MEM I nutrient system, including insulin, transferrin, hypoxanthine, thymidine, and trace elements. The formulation differs from the conventional phenol-red-containing Opti-MEM I medium by omitting that indicator dye. The 1× liquid medium is supplied as a 500 mL bottle and can support reduced-serum culture of mammalian cells such as CHO, HEK, Sp2, and BHK-21. It is also suitable for preparing lipid-based transfection complexes. The medium itself contains no fetal bovine serum; serum supplementation for sustained culture depends on cell type and growth requirements. A sodium bicarbonate buffering system requires a CO2-controlled environment for maintaining physiological pH. Store the bottle at 2–8 °C away from light, without freezing. The shelf life is 12 months from the date of manufacture, different from the phenol-red-containing formulation.
