Description

The HM47 monoclonal antibody specifically binds to the cytoplasmic domain of CD79a. CD79a is a 47 kDa type 1 transmembrane glycoprotein present on B lymphocytes. CD79a is also referred to as mb-1, IGA and Ig-alpha (Igα). It is expressed on B cells at various stages of differentiation, from pre-B cell stage, probably before expression of cytoplasmic µ chain, to plasma cell stage, in which it is detected only in the cytoplasm. CD79a associates with CD79b to form part of the B-cell receptor complex. It has been suggested that CD79a may play a role in mediating the transport of IgM to the cell surface. This antibody has been found to crossreact with permeabilized A20 cells (mouse B cell line).

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The antibody was conjugated with BD Horizon BV421 under optimum conditions, and unconjugated antibody and free BD Horizon BV421 were removed.
1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
2. Source of all serum proteins is from USDA inspected abattoirs located in the United States.
3. An isotype control should be used at the same concentration as the antibody of interest.

5. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

Application Note

BD Horizon BV421 Mouse Anti-Human CD79a is an intracellular flow cytometry antibody targeting the cytoplasmic domain of CD79a. Clone HM47 detects the Ig-alpha component of the B-cell receptor signaling complex, which associates with CD79b. CD79a is present during multiple stages of B-cell differentiation and supports phenotyping of B-lineage populations when combined with surface markers. Because the HM47 epitope lies inside the cell, fixation and permeabilization are necessary before antibody staining; routine live-cell surface staining alone will not expose the target. Brilliant Violet 421 is efficiently excited by a 405 nm violet laser and detected in a violet/blue fluorescence channel around 421 nm. The vial contains 50 tests, with 5 µL per test. Include permeabilized negative controls and appropriate compensation or spectral unmixing controls. Store refrigerated, protected from light, and never freeze.
