Answer:Intracellular staining of human samples by flow cytometry is routinely tested.
Answer:Use 5 µL per test; a typical test contains 1 × 10^6 cells in 100 µL.
Answer:Store it undiluted at 4 °C, protect it from prolonged light exposure, and do not freeze it.
Alexa Fluor® 647 Mouse Anti-Human S100A8/A9 (MRP-8/14), Clone 5.5 is used for intracellular flow-cytometric detection of human S100A8/A9, also known as MRP-8/MRP-14 or calprotectin. The monoclonal antibody uses mouse BALB/c IgG1, κ and was generated against acute monocytic leukemia cells and monocytes. Human reactivity receives quality-control testing, and intracellular staining by flow cytometry is routinely tested. Affinity purification isolates the antibody from tissue-culture supernatant or ascites, and direct conjugation to Alexa Fluor 647 eliminates a separate fluorescent secondary-antibody step. Unreacted dye is removed after conjugation. The reagent arrives prediluted at a working volume of 5 µL per test; a typical test contains 1 × 10^6 cells in a 100 µL experimental sample. Alexa Fluor 647 emission can be collected with the same instrument settings used for allophycocyanin, which supports placement in APC-compatible detector channels during multicolor panel design. The formulation is an aqueous buffered solution containing BSA and no more than 0.09% sodium azide. Store the reagent undiluted at 4 °C, protect it from prolonged light exposure, and do not freeze it. Direct labeling, a defined per-test volume, human QC testing, and established APC-channel collection simplify incorporation into intracellular immunophenotyping panels.
