Description
The S5/1 monoclonal antibody specifically recognizes CD88 which is also known as C5a receptor (C5aR), Complement C5a receptor 1 (C5AR1) or C5a anaphylatoxin chemotactic receptor 1. CD88 (C5aR) is an ~43 kDa G type protein-coupled receptor with seven transmembrane domains encoded by C5AR1 (complement C5a receptor 1) which belongs to the rhodopsin superfamily. This receptor is variably expressed on neutrophils, basophils, eosinophils, monocytes, macrophages, dendritic cells, mast cells as well as human cell lines of myeloid origin, and certain cell types in several organs, including liver hepatocytes, lung bronchial and alveolar epithelial cells, lung vascular smooth muscle, endothelial cells, astrocytes, microglia, and fibroblast-like cells of the brain. The binding of the anaphylatoxin ligands C5a or C5a(desArg) to CD88 (C5aR) leads to various cellular responses including the shedding of selectins, upregulation of adhesion molecules, chemotaxis, granule exocytosis, activation of NADPH oxidase, and superoxide anion production. The S5/1 monoclonal antibody reportedly blocks anaphylatoxin ligand binding to CD88 (C5aR).
Bead-based compensation or unmixing controls, such as BD® CompBeads or BD™ SpectraComp™, can be used as surrogates to assess fluorescence spillover when bound to fluorochrome-conjugated antibodies. Although these beads have spectral properties similar to cells, variations in spectral emission may occur, resulting in differing spillover values compared to biological controls. Therefore, it is considered best practice to compare the spillover obtained from cells and bead-based controls when using BD® CompBeads or BD™ SpectraComp™ for the first time, to ensure they are appropriate for the intended application.
The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to the dye under optimum conditions and unreacted dye was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

2. Alexa Fluor® 647 fluorochrome emission is collected at the same instrument settings as for allophycocyanin (APC).
3. Source of all serum proteins is from USDA inspected abattoirs located in the United States.
4. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
5. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
6. For fluorochrome spectra and suitable instrument settings, please refer to our Multicolor Flow Cytometry web page at www.bdbiosciences.com/colors.
7. An isotype control should be used at the same concentration as the antibody of interest.
