Answer:Immobilized metal-ion affinity chromatography (IMAC).
Answer:No. Its iminodiacetic acid groups can be charged with a selected compatible metal ion.
Answer:Five columns, each containing 5 mL of chelating resin.
The Cytiva HiTrap Chelating HP columns contain Chelating Sepharose High Performance agarose resin with immobilized iminodiacetic acid chelating groups for metal-ion affinity chromatography. The resin can be charged with an appropriate metal ion, such as nickel or cobalt, to capture compatible histidine-tagged proteins during recombinant protein purification. The highly cross-linked agarose matrix has an average bead size of approximately 34 µm and supplies 5 mL of packed media in each HiTrap column. Each pack includes five ready-to-charge columns for small-scale IMAC purification and method optimization. Unlike a fixed-metal affinity column, the chelating medium permits selection of the immobilized metal ion before use.
