Answer:Strong cation exchange chromatography, using negatively charged ligands to bind appropriately charged proteins.
Answer:One liter of bulk chromatography resin for packing a suitable column.
Answer:Elution can be achieved by raising salt concentration or adjusting buffer conditions, with the method optimized for the target protein.
Cytiva Capto S ImpAct is a strong cation exchange resin supplied as a 1 L package of loose chromatography medium for preparative protein purification. Its negatively charged sulfonate functional groups bind suitable positively charged biomolecules under conditions selected for the target's isoelectric point and buffer composition. The resin supports capture or intermediate purification steps where resolution, process throughput and impurity removal are important. Develop a method by selecting loading pH and conductivity, evaluating breakthrough capacity, and eluting bound material with an appropriate salt gradient or step. Because binding depends on the sample, neither an individual protein's capacity nor recovery should be assumed from resin family data alone. Pack the resin into compatible process hardware and check bed consolidation, pressure–flow characteristics and separation efficiency before production use. Establish cleaning-in-place and storage procedures that preserve resin lifetime and meet process requirements. This product is bulk resin, not a ready-to-use prepacked column.
