HisTrap FF 5 mL columns provide a ready-to-use immobilized nickel affinity capture step for histidine-tagged recombinant proteins. Each column contains 5 mL of precharged Ni Sepharose 6 Fast Flow resin in a polypropylene housing, and the commercial pack contains five columns. The greater resin bed volume supports higher total sample loading than a 1 mL column when operating conditions and binding capacity are suitable. Protein capture normally uses a compatible binding buffer, followed by washing and controlled elution with imidazole. Define loading conditions using the target protein and sample composition, and evaluate eluted fractions for purity and activity. When using ÄKTA chromatography systems, apply the column-specific flow and pressure limitations.
