Answer:It measures the formation of soluble formazan from cellular reducing activity, which serves as an indirect indicator of viable-cell metabolic activity.
Answer:No. Its colored formazan product is water soluble, so an MTT-style crystal-solubilization step is unnecessary.
Answer:Background absorbance, redox-active compounds and differences in incubation time can change absorbance independently of cell proliferation.
Roche Cell Proliferation Reagent WST-1 is a ready-to-use tetrazolium-based reagent for measuring metabolic activity in viable cultured cells. Mitochondrial and other cellular reductase activity converts WST-1 into a soluble colored formazan product, enabling an absorbance-based readout in microplate experiments. This 25 mL liquid format supports cell proliferation, cytotoxicity and viability studies without the insoluble-crystal solubilization step used by traditional MTT assays. The measured signal reflects cellular reducing activity rather than a direct count of living cells. Establish a working range with appropriate cell seeding densities, incubation periods, medium blanks and untreated controls. Test compounds that affect redox activity or contribute optical absorbance can interfere with interpretation, even when cell number remains unchanged. Measurements are commonly collected near 450 nm with a reference wavelength as appropriate for the plate reader. Consistent culture conditions are essential for comparisons across treatments.
