Answer:No. DNA binds to a pretreated silica-particle suspension; separation is performed with centrifugation and washing.
Answer:The reagent set contains silica matrix and accompanying buffers for 100 standard gel extraction reactions.
Answer:No. Add 80 mL absolute ethanol to the supplied wash-buffer concentrate before its first use.
The Roche Agarose Gel DNA Extraction Kit isolates DNA fragments excised from TAE- or TBE-based agarose gels after electrophoretic separation. This 100-reaction kit uses pretreated spherical silica matrix in suspension rather than a disposable silica spin-column cartridge. A chaotropic agarose-solubilization buffer releases DNA from the gel and promotes adsorption onto the silica particles. After centrifugation and successive washes, the DNA is released in low-salt elution buffer or water for downstream cloning, restriction digestion or labeling. The silica suspension, agarose solubilization buffer, nucleic-acid binding buffer and concentrated wash buffer are supplied as separate components. Add the specified absolute ethanol to the wash buffer before first use and use an appropriate gel slice-to-buffer ratio. Fragment recovery depends on length, gel composition, completeness of dissolution and elimination of wash-buffer carryover. Avoid carrying silica particles into the final eluate, as this can compromise subsequent enzymatic reactions. Store at 15–25°C as specified for the kit.
