PowerQuant® System is used for human DNA quantification and quality assessment before forensic STR amplification. This five-dye, four-target hydrolysis-probe qPCR assay measures amplifiable autosomal DNA and Y-chromosomal DNA in a single reaction while also evaluating DNA degradation and detecting PCR inhibition through an internal control. The 200-reaction configuration includes a male genomic DNA standard, primer/probe/internal-control mix, 2X master mix, amplification-grade water, and dilution buffer. The system is designed to detect very low levels of human DNA and reports sensitivity down to 0.5 pg/µL under the specified workflow. Its combined quantity, male-DNA, degradation, and inhibition information helps analysts choose an appropriate downstream STR strategy and identify extracts that may need dilution or other intervention. The assay is compatible with supported real-time PCR instruments and can be incorporated into automated liquid-handling workflows used in forensic laboratories.
