Advanced DMEM requires supplementation with 1–5% Fetal Bovine Serum and 4 mM L-glutamine or GlutaMAX™ supplement. Many cell lines do not require adaptation to this media. The FBS concentration must be optimized for each cell line to obtain maximum serum reduction. Advanced DMEM uses a sodium bicarbonate buffer system (3.7 g/L), and therefore requires a 5–10% CO_2 environment to maintain physiological pH.

Application Note

Gibco Advanced DMEM is a ready-to-use liquid basal medium for mammalian cell culture under reduced serum supplementation. This configuration contains ten 500 mL bottles and is formulated with high glucose, sodium pyruvate, nonessential amino acids and phenol red, but without L-glutamine. Its specialized supplements, including insulin, transferrin and other nutrient components, support reduced fetal bovine serum conditions in compatible cell lines; the suitable serum reduction must be optimized for the cells and workflow. Add L-glutamine or GlutaMAX at the recommended concentration and supplement with serum before culture following the validated protocol. A common starting recommendation is 1–5% FBS with 4 mM glutamine or GlutaMAX. Advanced DMEM uses sodium bicarbonate buffering and requires an appropriate CO₂ atmosphere for physiological pH control. The sterile-filtered medium is for research use and should be stored protected from light at 2–8 °C.
