Answer:KM71H is a MutS strain with slow methanol utilization.
Answer:The strain carries an aox1::ARG4 disruption together with arg4.
Answer:This configuration is supplied as one stab culture.
KM71H Pichia pastoris Yeast Strain is used for recombinant protein expression when a MutS methanol-utilization phenotype is desired. The strain carries the genotype aox1::ARG4, arg4 and has a MutS His+ phenotype. Disruption of AOX1 means that methanol is metabolized more slowly than in Mut+ strains, so transformed colonies do not require additional screening to identify the MutS phenotype. KM71H is compatible with Zeocin-resistant Pichia expression vectors and can be useful when slower methanol utilization improves expression behavior for a particular recombinant protein. The strain is supplied as one stab culture for research use. It provides a defined alternative to X-33 or GS115 for comparing expression level, protein folding, secretion and fermentation behavior across different Pichia host phenotypes.
