Answer:The random hexamer mixture is supplied as 120 µL at 0.2 µg/µL, totaling 24 µg.
Answer:Yes. Random hexamers can initiate cDNA synthesis along many RNA sequences without requiring a poly(A) tail.
Answer:Store the primer at −20 °C under nuclease-free handling conditions.
Thermo Scientific Random Hexamer Primer is a mixture of random six-nucleotide oligonucleotides that initiates reverse transcription at multiple locations along an RNA template. The primer population has free 5′ and 3′ hydroxyl ends and supports preparation of cDNA from total RNA for downstream RT-PCR, RT-qPCR and transcript analysis. The liquid formulation is supplied in a 120 µL volume at 0.2 µg/µL, corresponding to a total of 24 µg. Distribute primer and RNA into nuclease-free reaction vessels, then combine with an appropriate reverse transcriptase, dNTP mixture and reaction buffer. Random priming can generate cDNA from RNA molecules without poly(A) tails and from regions where oligo(dT) priming would be unsuitable, although transcript coverage and background depend on template integrity and experimental design. Store the primer frozen at −20 °C and minimize unnecessary thawing. Treat the reagent as a reverse-transcription primer rather than a gene-specific PCR amplification primer.
