The Tosoh TSKgel Phenyl-5PW hydrophobic interaction chromatography column contains 10 µm polymethacrylate particles bearing phenyl functional groups. The analytical column is 7.5 mm in internal diameter and 75 mm long, with a 100 nm pore size that helps large biomolecules access the separation surface. Hydrophobic interaction chromatography separates proteins and related biomolecules based on their surface hydrophobicity under salt-dependent binding conditions. Samples are commonly loaded in a suitable higher-ionic-strength buffer and eluted by reducing salt concentration, with conditions selected to protect molecular integrity and recovery. Phenyl-5PW offers a relatively hydrophobic selectivity within the Tosoh HIC column family and is useful for screening monoclonal antibodies, fragments and aggregate-related differences. Equilibrate thoroughly before sample loading, filter samples to limit particulate fouling, and evaluate recovery and resolution with representative material. This column is not a reversed-phase C18 HPLC column; its retention mechanism and typical mobile phases are different.
