CM Sephadex C-50 is a weak cation-exchange medium made from cross-linked dextran with carboxymethyl functional groups. The dry resin is hydrated before use and can be applied in column chromatography or batch ion-exchange procedures for proteins, peptides and other charged biomolecules. Binding is influenced by buffer pH, ionic strength and the protein's net charge, so preliminary tests should establish equilibration and elution conditions for the sample. Batch processing can be useful when the target application prioritizes capacity and simple handling over high-resolution gradient separation. Unlike strong sulfonate-based exchangers, carboxymethyl groups have pH-dependent ionization, which matters in method development.
