Cytiva PD-10 desalting columns are prepacked gravity-flow gel filtration devices used to remove salts and other low-molecular-weight substances from protein samples. Each column contains approximately 8.3 mL of Sephadex G-25 Medium resin, with a recommended sample volume of 1.0–2.5 mL for the gravity-flow procedure. Thirty columns are included in the package. The porous gel separates macromolecules from smaller dissolved species because proteins elute ahead of salts and other molecules that enter the Sephadex pores. PD-10 columns are useful for buffer exchange, desalting, removing free labels or preparing purified protein solutions for subsequent assays and chromatography. Use a suitable equilibration buffer and collect the appropriate fraction for the selected protocol. The columns are disposable preparation tools rather than ion-exchange columns or cartridges for affinity capture. Sample recovery, dilution and separation efficiency depend on loading volume and correct column handling, so results should be assessed against the downstream analytical requirements.
