Answer:The typical fractionation range is approximately 10,000 to 600,000 Da for globular proteins.
Answer:Size-exclusion chromatography separates molecules by their access to pores in the resin matrix; larger species generally elute earlier than smaller ones.
Answer:No. It is bulk chromatography resin intended for packing an appropriate preparative column.
Superdex 200 Prep Grade is a preparative size-exclusion chromatography medium for separating proteins and protein complexes on the basis of their effective hydrodynamic size. Its composite cross-linked agarose and dextran matrix provides a fractionation range of approximately 10,000–600,000 Da for globular proteins. The 150 mL resin pack enables preparation of columns for aggregate separation, buffer exchange-related fractionation and final protein polishing where molecular size is the principal separation mechanism. For effective resolution, pack a uniform bed, use a compatible isocratic mobile phase and keep injection volume appropriate to column geometry. Sample concentration, viscosity and interactions with the matrix can influence peak shape and recovery. Size-exclusion retention is not a direct measurement of molecular weight without calibration, and resin volume alone does not specify the size of a finished chromatography column.
