Answer:Hydrophobic interaction chromatography with butyl ligands on a cross-linked agarose matrix.
Answer:One liter of bulk chromatography medium.
Answer:By decreasing the salt concentration after high-salt binding.
Butyl Sepharose High Performance is a hydrophobic interaction chromatography medium for the purification and polishing of biomolecules based on surface hydrophobicity. Butyl ligands are coupled to a highly cross-linked agarose matrix to support protein capture or resolution under high-salt binding conditions followed by decreasing-salt elution. The High Performance bead structure enables high-resolution HIC separations in packed chromatography columns. The 1 L bulk resin format is used in method development, scale-up, and production purification where protein hydrophobicity provides useful selectivity after preceding chromatographic steps. It is a chromatography medium, not a ready-to-run prepacked column.
