Answer:Confirmation and presumptive identification of intestinal enterococci in water testing.
Answer:Esculin hydrolysis produces esculetin, which reacts with ferric ions to form a dark complex.
Answer:It is a 500 g bottle of non-sterile granulated medium.
Bile Aesculin Azide Agar is designed for selective culture and presumptive identification of intestinal enterococci during microbiological water analysis. The GranuCult® plus dehydrated medium is supplied as 500 g of granules and is suitable for the confirmation stage of an ISO 7899-2 membrane-filtration workflow. Bile salts help suppress susceptible accompanying organisms, and sodium azide suppresses Gram-negative microbial flora. Enterococci hydrolyze esculin to esculetin; the resulting reaction with ferric ions produces a dark olive-green to black complex around typical colonies. This reaction, together with bile tolerance, supports identification of the presumptive colonies recovered from a primary selective membrane agar. The recommended preparation is approximately 54.85 g per liter of water, with a final pH of 6.9–7.3 after autoclaving under the stated conditions. The medium is supplied non-sterile and requires preparation before use. Store the tightly closed container dry, protected from light, at 15–25 °C. It is a confirmation medium rather than the TTC-containing primary enumeration medium.
