Answer:Total dietary fiber is determined by enzymatic digestion of nonfiber components followed by gravimetric measurement corrected for protein and ash.
Answer:It includes heat-stable α-amylase, protease, amyloglucosidase and Celite filtration aid.
Answer:The kit supplies materials for approximately 100 assays and is stored at 2–8°C.
Merck Total Dietary Fiber Assay Kit determines dietary fiber in food and related analytical samples by an enzymatic-gravimetric method based on AOAC 985.29. Heat-stable α-amylase gelatinizes and digests starch, followed by protease treatment to remove proteins and amyloglucosidase digestion of residual starch fragments. Ethanol precipitates soluble dietary fiber so that the insoluble and precipitated soluble fractions can be collected on a filtration medium. After ethanol and acetone washes, the retained residue is dried and weighed. Separate aliquots undergo protein and ash determination, and the corrected residue mass is used to calculate total dietary fiber. The kit contains heat-stable α-amylase (10 mL), amyloglucosidase (10 mL), protease (500 mg) and Celite (50 g), sufficient for approximately 100 assays. The method also requires appropriate solvents, apparatus and reagents for protein and ash determination. Store the kit at 2–8°C and perform blank and control measurements with each analytical series.
