Answer:Successive dilution of fluorescent labeling as stained cells divide, measured by flow cytometry.
Answer:Excitation near 409 nm and collection through a 450/50 nm band-pass filter.
Answer:A 500 µg vial of lyophilized fluorescent dye for reconstitution before cell labeling.
The Invitrogen eBioscience Cell Proliferation Dye eFluor 450 is an amine-reactive violet fluorescent tracer for following cell divisions by flow cytometry. The fluorescent label covalently binds to proteins containing primary amines and becomes distributed between daughter cells during proliferation, creating successive peaks of decreasing signal intensity. Its excitation maximum is approximately 409 nm, with detection using a 450/50 nm band-pass filter. The 500 µg vial contains lyophilized dye that can be reconstituted in anhydrous DMSO to prepare a labeling stock. The dye supports monitoring of up to seven cell generations under suitable assay conditions and can be used with downstream fixation and permeabilization workflows.
