Rapid DNA Ligation Kit provides T4 DNA ligase and a specially formulated 5X buffer for fast joining of sticky-ended or blunt-ended DNA. This configuration supports 150 reactions. The optimized chemistry can complete routine ligation in about 5 minutes at room temperature, reducing the incubation time required for many cloning workflows while maintaining ligation efficiency comparable with longer standard reactions. The reaction mixture can be used directly for conventional bacterial transformation procedures, which simplifies cloning workflows that move immediately from ligation to transformation. The kit is suitable for routine cloning, blunt-end cloning, library construction, and TA cloning. Nuclease-free water is supplied with the ligase and buffer. If the ligation mixture will be used for electroporation, the ligase should be removed or inactivated using a suitable cleanup procedure before the electric pulse.
