Answer:The solution contains 25 mM magnesium chloride and is supplied as four 1.25 mL tubes.
Answer:Magnesium concentration affects polymerase activity, amplification yield and specificity; it is optimized against the complete reaction composition.
Answer:No. It supplies magnesium chloride only and must be combined with the other required reaction components.
Thermo Scientific 25 mM magnesium chloride solution provides a defined magnesium-ion source for adjusting PCR and other magnesium-dependent enzymatic reactions. Magnesium ions influence DNA polymerase activity, primer-template interactions and the balance between amplification yield and specificity. The aqueous reagent is passed through a 0.22 µm membrane and supplied in four 1.25 mL aliquots. It can be used to optimize reactions assembled with compatible polymerases when their magnesium requirement differs from the baseline reaction-buffer composition. Calculate final free and total magnesium concentrations from all reaction inputs, including magnesium already present in the buffer and chelating effects from nucleotides or additives. Optimize using a controlled concentration series while keeping other reaction parameters stable. Do not confuse this magnesium supplement with a complete PCR buffer or a ready-made PCR master mix. Maintain nuclease-free technique, avoid contaminating stock tubes and store the reagent at −20 °C in a non-frost-free freezer.
